چکیده

RNA replication of Cymbidium ringspot virus (CymRSV) normally occurs in plant and yeast cells in association with membranes derived from peroxisomes. However, in yeast strains devoid of these organelles CymRSV RNA replication takes place on the perinuclear membrane and cytoplasmic endoplasmic reticulum (ER) strands. The association of the CymRSV RNA replication complex to ER is mediated by the ORF 1encoded protein p33. To analyze which part of this protein contains the information leading to localization to ER, a deletion analysis was carried out. The behaviour of deletion mutants was studied by confocal immunofluorescence microscopy and membrane floatation gradient analysis. It was shown that the signal targeting p33 to ER membranes largely coincides with the peroxisomal targeting signal, but contains additional elements in the first 87 amino acids.

Cymbidium ringspot virus defective interfering RNA replication in yeast cells occurs on endoplasmic reticulum-derived membranes in the absence of peroxisomes.

The replication of Cymbidium ringspot virus (CymRSV) defective interfering (DI) RNA in cells of the yeast Saccharomyces cerevisiae normally takes place in association with the peroxisomal membrane, thus paralleling the replication events in infected plant cells. However, previous results with a peroxisome-deficient mutant strain of yeast had suggested that the presence of peroxisomes is not a s...

Cytological analysis of Saccharomyces cerevisiae cells supporting cymbidium ringspot virus defective interfering RNA replication.

The replicase proteins p33 and p92 of Cymbidium ringspot virus (CymRSV) were found to support the replication of defective interfering (DI) RNA in Saccharomyces cerevisiae cells. Two yeast strains were used, differing in the biogenesis of peroxisomes, the organelles supplying the membranous vesicular environment in which CymRSV RNA replication takes place in infected plant cells. Double-labelle...

Cymbidium ringspot virus harnesses RNA silencing to control the accumulation of virus parasite satellite RNA.

Cymbidium ringspot virus (CymRSV) satellite RNA (satRNA) is a parasitic subviral RNA replicon that replicates and accumulates at the cost of its helper virus. This 621-nucleotide (nt) satRNA species has no sequence similarity to the helper virus, except for a 51-nt-long region termed the helper-satellite homology (HSH) region, which is essential for satRNA replication. We show that the accumula...

In situ characterization of Cymbidium Ringspot Tombusvirus infection-induced posttranscriptional gene silencing in Nicotiana benthamiana.

In plants, posttranscriptional gene silencing (PTGS) is an ancient and effective defense mechanism against viral infection. A number of viruses encode proteins that suppress virus-activated PTGS. The p19 protein of tombusviruses is a potent PTGS suppressor which interferes with the onset of PTGS-generated systemic signaling and is not required for viral replication or for viral movement in Nico...

Molecular mechanism of RNA silencing suppression mediated by p19 protein of tombusviruses.

RNA silencing is an evolutionarily conserved surveillance system that occurs in a broad range of eukaryotic organisms. In plants, RNA silencing acts as an antiviral system; thus, successful virus infection requires suppression of gene silencing. A number of viral suppressors have been identified so far; however, the molecular bases of silencing suppression are still poorly understood. Here we s...
